Preparation - frozen tissue

Female C57BL/6 or SJL mice 8 to 12 weeks of age are acclimated to our facility for at least 7 days.

In C57BL/6 mice, EAE is induced according to the protocol EAE Induction by Active Immunization in C57BL/6 Mice.

In SJL mice, EAE is induced according to the protocol EAE Induction by Active Immunization in SJL Mice.

Mice are scored daily for EAE severity on the scale of 0-5 in 0.5 unit increments (see protocol), and weighed 3 times/week (Monday, Wednesday, Friday).

Mice are euthanized at the appropriate stage of disease, immediately perfused with PBS, then spleen & spinal cord are isolated and snap frozen in liquid nitrogen.

Tissues are stored at -80 °C until shipment.

Preparation - fresh cells

Female mice of age 8 to 14 weeks are acclimated to our facility for at least 7 days, then immunized with antigen subcutaneously in the lower back, close to the base of tail, as follows:

11 days later (10 days for SJL mice), mice are euthanized and cell suspension(s) from spleen and/or inguinal lymph nodes are prepared.

Red blood cells in spleen are lysed.

Cells are resuspended in complete tissue culture media and cell concentrations determined and marked on tubes.

Cells are shipped the same day for overnight delivery.

Detailed contents - frozen tissue

Each product unit contains the entire spleen and spinal cord tissue from a single mouse.

Each organ is supplied in a separate 1.5 mL cryogenic tube. Tubes are marked with a mouse ID and tissue weight. (Typical spinal cords are 80 to 100 mg each, spleens are typically 100 to 200 mg.)

The daily EAE severity scores and weight history for the mouse are provided for each mouse in a spreadsheet, via email.

Detailed contents - fresh cells

Centrifuge tubes (15 or 50 mL) with cells in complete tissue culture media (90% RPMI 1640, 10% fetal calf serum, supplemented with 0.1 mM MEM non-essential amino acids, 1 mM MEM sodium pyruvate, 2 mM L-glutamine, 100 UI/mL penicillin, 100 µg/mL streptomycin, 5 x 10-5 M 2-mercaptoethanol).

Each cell type is supplied in a separate centrifuge tube, together with cell counts (cells/mL) for the cell type. If cells from multiple mice (of the same catalog number) are ordered, each type of cells will be pooled in one tube with one cell count.

Each product unit contains the entire suspension of spleen and/or lymph node cells from one mouse.
